I remember a few years back the guys at vg put out a conference call in which they discussed labbing (I think it was the first conference call they ever did).
One of the main things they said was that there was really no WRONG way to lab, but there are ways of labbing that are efficient.
One of the main ways to lab is to take 2 controllers and pick your play...whether it be offense if you want to work on that or defense....
pick a random play to go against and just go from there.
If your offense, try to nail down all your reads and the order in which you should be looking. Try to guess what type of defense it is presnap and then figure out where you should go. once the ball is snapped see if you're presnap was right. If it was hit your first option, if it wasn't and the first option is covered, move onto your second option and so on
defensively, what I like to do is call one defense on the computer (All madden of course) and let the computer tear it up...this way I learn the defensive assignments and who I can playmaker, who I should control etc.
Bottom line...when you're labbing your stuff, try to learn all the different variations to your plays and do it against multiple defense and offense so that when you see another human, you're prepared
The secret to success? success = preparation plus opportunity. when you're presented with an opportunity, if you've thorougly prepared yourself, you'll be successful
Hope this helps
